The long term goal of this research program is to identify and develop novel antimicrobial agents that target one of two highly conserved cellular functions, bacterial translation or peptidoglycan (PG) synthesis, to treat resistant bacteria. Translation, the process by which mRNA is used to produce proteins, is essential for bacterial survival. Each mRNAs corresponds to a specific protein and consists of a 5’end, a 3’end with a conserved sequence of A’s, T’s, C’s, G’s, and U’s, and a center of non-coding sequences including the ribosome binding site. For each translational event, ribosomes translate both the mRNA 5’end and 3’end sequentially to produce a polypeptide chain that is linked through a peptide bond. PG, a polymer made up of glycan chains cross-linked with peptides, is the major component of the cell wall of bacteria. It is synthesized on the cytoplasmic side of the plasma membrane. The 5’end of the majority of bacterial mRNAs is translationally coupled to PG synthesis, and not surprisingly, most of the enzymes involved in PG synthesis have been found to form part of the translation machinery. The objective of this proposal is to gain a molecular understanding of the interaction between translation and PG synthesis, and use this information to discover novel antibiotics that target the ribosome or PG synthesis. In our first aim, we will use genetic and biochemical assays to elucidate the relationship between translational components and PG synthesis. The second aim of this research proposal involves the identification and characterization of antibiotics that target translation. The tool is not an interface per se and so, pressing the keys to indicate what to test are shown in the window below.
The Help menu allows for a preview of all key binds and commands that you can use.

It goes without saying that the test is done in full screen and there is nothing else on the computer screen than the testing window. Once you are done with checking, simply press the ESC key to exit.

After performing a short and non-scientific analysis of pixcheck on our end, we have concluded that the application could have been much more user friendly. The Help menu does not provide a link to a documentation that might have been very useful in understanding the commands and key binds.
Adding a completion menu to the file that pops up when you type a command may make the utility more user friendly. The user interface, even if basic and not very pleasing, is efficient and it also provides a clear presentation of the results of the test.
At times, the results can be a bit confusing because there are listed as good or bad pixels. Furthermore, pixcheck does not seem to be smart in choosing the positions to test.
When you launch the program, the color circle pops up and you can use your keyboard to navigate the colors. The program also offers two types of tests, the standard test and the single color test. With the single color test, you can quickly identify whether the problem is with the red, green, or blue.
The major drawback of pixcheck is that it does not collect the results but simply displays them. So, there is no way to do a report of the error found in a specific pixel and to create a copy of the list of bad pixels.
Sometimes, it may be very tricky to recognize which pixel on the screen is a bad one. When you hit a pixel that seems like a good one, it may still be a bad one. By using the automatic selection
