Silymarin was incorporated into solid lipid nanoparticles (SLNs) with the aim of improving its dissolution and bioavailability. The batch of nanoparticles was prepared using glyceryl monostearate (GMS) as the lipid material, and polyvinylpyrrolidone (PVP) as the stabilizing polymer, and loaded with 10% (w/w) of silymarin. The particle size, zeta potential, thermal analysis, dissolution, and in vitro release of the SLNs were investigated. The particle size was in the range of 153-279 nm with a narrow size distribution. The average zeta potential was -38.2 mV. The thermal analysis revealed that GMS melted around 72 degrees C, while PVP remained stable up to 100 degrees C. There was a rapid release from the SLNs during the initial 5 min, followed by a slow release. After 8 h the drug release was 84% from SLNs. There was no significant difference in the bioavailability of the silymarin from the SLNs as compared to the silymarin suspension (p > 0.05). The data obtained from the isothermal titration calorimetry (ITC) study revealed that the silymarin-SLNs complex formation was an enthalpy-driven process. This study indicates that the solid lipid nanoparticles can be used to encapsulate poorly water-soluble drugs to improve their oral bioavailability.Socioeconomic factors and risk of Alzheimer’s disease in the elderly.

A total of 5,228 individuals from the same community who were assessed for dementia at the study site over a 10-year period were divided into AD and other dementias.
